I'd already suspected that cause I read about that in the past and it also can be read in the location settings section too. I just asked myself if there could be other purposes beyond that. WiFi-/Bluetooth-Search and Network Location are disabled too, so these should be excluded.

Do you mean the observed connections in the DNSNet app. No, I'm convinced, these are definitely triggered by the App. There are connections only logged (and counted) when they are actually triggered. As I described, they count, when tab the start button for the blocking function within the app. I can't imagine that the system checks the the ability of providing location for emergency calls by chance exactly when you tap this button. And surely not e.g. 20 times in 10 seconds (when you tap the button 20 times in 10 seconds). Same with the ntp.org connections, they are triggered together with supl exactly when tapping the start button or toggling on logging. (Or maybe toggling off logging, can't confirm that exactly while it can only be seen after logging turned o again.)
Ok, there's still the chance that it's a bug within the app showing these mistakenly. But any other connections seem to be shown correctly. The only exception I could realize is time.grapheneos.org, which won't be logged there even when time synchronization really happens (and also no other connection instead). So I still wonder why these connections are triggered by the app.
GrapheneOS Yes, but Samsung gnssd introduced another NTP connection we need to deal with by changing it to time.grapheneos.org as we did with Qualcomm previously.
That is already changed to time.grapheneos.org? Or if not, it's not using pool.ntp.org?
